They were indeed! Doc Ock was actually the original founder who got the group together and Mysterio was one of the original members. So they've worked together but they've also turned on each other so Beck may be somewhat wary of Otto.

But I'd love for them to interact! He is aware of multiverses too so it's easy for Otto to explain he's a different version than the one he knows.
